Canine Pancreatic Ribosomes
نویسنده
چکیده
Purified microsomal and postmicrosomal ribosomes have been prepared from dog pancreas by a method involving magnesium precipitation and treatment with sodium deoxycholate. The purified ribosomes were shown to be substantially free from contamination with supematant protein in an experiment employing a radioactive supematant fraction. They contain less than 0.1% of digestive enzymes known to be major products of synthesis by the gland. The purified particles appear to contain 60% RNA when pancreatic ribosomal protein is used as the protein standard in the Lowry procedure. Different values are obtained, however, when either bovine serum albumin or bovine pancreatic chymotrypsinogen is used as standard. The monoribosome behaves ultracentrifugally as an 80 S species in the presence of 0.1 M KC1 and as a 74 S species in its absence. Lyophilization causes partial dissociation of the monoribosome to ribosomal subunits, complete dissociation being effected by ethylenediaminetetraacetate at a concentration of 2 pmoles per mg of ribosome. The large and small subunits sediment, respectively, as 50 S and 33 S species in the presence of 0.1 M KC1 and as 34 S and 29 S species in the absence of KCl. The individual subunits have been isolated by density gradient centrifugation, and their chemical composition has been studied.

Reduced Pancreatic Exocrine Function and Organellar Disarray in a Canine Model of Acute Pancreatitis
The aim of the present study was to investigate the pancreatic exocrine function in a canine model and to analyze the changes in organelles of pancreatic acinar cells during the early stage of acute pancreatitis (AP). AP was induced by retrograde injection of 5% sodium taurocholate (0.5 ml/kg) into the main pancreatic duct of dogs. The induction of AP resulted in serum hyperamylasemia and a mar...
متن کاملSubcellular distribution of calcium-binding proteins and a calcium- ATPase in canine pancreas [published erratum appears in J Cell Biol 1990 Oct;111(4):1726]
Using a 45Ca blot-overlay assay, we monitored the subcellular fractionation pattern of several Ca binding proteins of apparent molecular masses 94, 61, and 59 kD. These proteins also appeared to stain blue with "Stains-All." Additionally, using a monoclonal antiserum raised against canine cardiac sarcoplasmic reticulum Ca-ATPase, we examined the subcellular distribution of a canine pancreatic 1...
متن کاملAn immunoassay for canine pancreatic elastase 1 as an indicator for exocrine pancreatic insufficiency in dogs.
The detection of pancreatic elastase 1 in stool samples has become the noninvasive gold standard for the diagnosis of pancreatic insufficiency in humans. Accordingly, the development of a sandwich-ELISA specific for canine pancreatic elastase 1, based on monoclonal antibodies, is presented here. The test has a detection range of 4-240 microg canine pancreatic elastase l/g feces. The intraassay ...
متن کاملMitochondrial and cytoplasmic ribosomes and their activity in blood and culture form Trypanosoma brucei
Ribosomes of Trypanosoma brucei, a parasitic, flagellated protozoan (order Kinetoplastida), were identified on sucrose density gradients by their radioactively labeled nascent peptides. Ultraviolet absorption revealed only cytoplasmic ribosomes which served as internal sedimentation markers. Synthesis on cytoplasmic ribosomes was completely inhibited by cycloheximide. In the presence of this an...
متن کاملEffects of post-irradiation incubation on the fibroin synthetic activity of silkgland ribosomes.
Isolated silkgland ribosomes were irradiated with X-rays, and their activities to synthesize fibroin in vitro were examined. A marked decrease in their activities was observed when ribosomes were incubated at 37•Ž following X-irradiation. Per cent decrease in the ribosomal activity leveled off at 37•Ž after 30 minutes incubation . The extent of decrease was dependent on X-ray dose. Treatment of...
